This 2022 paper is a phytochemical study that isolated constituents from twigs and leaves of Tabernaemontana sananho and identified 12 alkaloids through physicochemical properties and spectroscopic analysis. Because discussion of sananga often asks which alkaloids the plant might contain, it is included as primary evidence grounded in specific plant parts and analytical methods.
The material analyzed was a laboratory-prepared fraction from twigs and leaves, not the sananga eye preparation generally described as made from roots or bark and water. Compounds present in twigs and leaves are not necessarily present in an eye preparation in the same composition or concentration, and isolation alone does not support inference about pharmacologic effects in humans.
Study design
The researchers used twigs and leaves of T. sananho as starting material and combined silica gel, ODS, and Sephadex LH-20 column chromatography, preparative HPLC, thin-layer chromatography, and other methods to isolate and purify constituents. Structures of the recovered compounds were identified from physicochemical properties and spectroscopic data.
This design separates a complex botanical mixture and compares it with data on known compounds to describe what was present. Ocular administration, comparison of activity in cells or animals, and human clinical assessment were outside the study purpose. The work also did not quantify contents or transfer into an aqueous preparation.
Content
The 12 identified compounds were coronaridine N-oxide, 3-oxocoronaridine, 3-oxo-7S-coronaridine hydroxyindolenine, tabersonine, tabersonine N-oxide, 3-oxotabersonine, 5β-hydroxy-3-oxotabersonine, lochnericine, (+)-voaphylline, deacetylakuammiline, harmine, and caffeine. This list demonstrates diversity of plant constituents, not their amounts or a list of actions in a finished eye preparation.
The authors reported coronaridine N-oxide as a new natural product; 5β-hydroxy-3-oxotabersonine and caffeine as isolated from the genus Tabernaemontana for the 1st time; and the other compounds as obtained from T. sananho for the 1st time. “1st” refers to reports of isolation in the literature, not that a constituent is always present or explains the effects of traditional use.
Plant chemistry can vary with genetics, growing conditions, collection time, plant part, drying, and extraction. Laboratory separation involving organic solvents also differs in extraction selectivity from a traditional water preparation. Connecting this paper to sananga quality assessment would require separate quantitative analysis of actual product batches.
Results
The direct result was isolation and identification of 12 alkaloids from the twig-and-leaf sample, adding new phytochemical records for several constituents. This provides a concrete account of the chemical diversity of T. sananho and a reference for future chemotaxonomy, quality analysis, and biological-activity research.
The result does not establish that sananga eye drops contain all 12 compounds, that each reaches the eye, or that they affect vision, infection, or inflammation. Constituent identification is an entry point for mechanistic hypotheses, not a final determination of safety or clinical efficacy.
Limitations
The public bibliographic and abstract information is insufficient to fully assess sample batches, yield of each compound, quantitative concentration distributions, or details of biological-activity tests. A botanical sample from 1 study may not represent all T. sananho across regions or seasons. Because a DOI could not be confirmed in databases, this page does not assign an unverified identifier.
Findings from twigs and leaves cannot be extrapolated to roots, bark, latex, or an aqueous eye preparation. Even if separate research reports activity for an individual compound, results may differ with concentration in a mixture, metabolism, interactions, and route of administration. The study contains no human safety or efficacy data.
Safety
Identification of compounds is not evidence that a liquid potentially containing them is safe for the eye. Alkaloids may be bioactive, making them candidates for benefit or toxicity depending on concentration, exposure site, and metabolism. Eye irritation, corneal cytotoxicity, sterility, and adverse events were not assessed.
